Overview

Ilixadencel + sorafenib is a combination therapy investigated for the treatment of advanced hepatocellular carcinoma (HCC). Ilixadencel (formerly INT-001) is an off-the-shelf, cell-based immune primer consisting of activated allogeneic dendritic cells derived from healthy donors. When injected intratumorally, it is designed to recruit and activate the patient's own immune cells, leading to a systemic, tumor-specific T-cell response. Sorafenib (Nexavar) is an orally administered small molecule multi-kinase inhibitor that targets several tyrosine kinases involved in tumor growth and angiogenesis, including VEGFR-1, -2, -3, PDGFR-beta, and Raf kinases. While a Phase I/II trial (NCT01974661) demonstrated the safety and tolerability of the combination, clinical outcomes were limited, and preclinical data suggested that sorafenib's immunosuppressive potential might interfere with the immune-priming mechanism of ilixadencel.
