Illumen Therapeutics is developing a first-in-class small molecule inhibitor of Poly(A)-specific ribonuclease (PARN) to treat aggressive cancers. The therapeutic strategy focuses on restoring the function of the p53 tumor suppressor, often referred to as the 'Guardian of the Genome.' In many cancers, p53 is not mutated but is functionally suppressed or downregulated. PARN is an enzyme responsible for the deadenylation and subsequent degradation of various mRNA transcripts, including those in the p53 pathway. By inhibiting PARN, the drug aims to stabilize p53 mRNA and increase the levels of functional p53 protein, thereby re-engaging natural tumor-suppressive mechanisms such as cell cycle arrest and apoptosis in malignant cells.
