ILT4.4-1BB.DAP10 CIR-NK is an engineered Natural Killer (NK) cell therapy developed by NKILT Therapeutics. It utilizes a Chimeric ILT Receptor (CIR) consisting of the extracellular domain of the Immunoglobulin-Like Transcript 4 (ILT4) receptor fused to intracellular costimulatory domains from 4-1BB and DAP10. This construct is designed to target tumor cells expressing Human Leukocyte Antigen G (HLA-G), an immune checkpoint often upregulated in cancers to evade immune surveillance. By binding HLA-G, the CIR-NK cells trigger activation and tumor cell lysis rather than the typical immunosuppression associated with HLA-G/ILT4 interaction. The cells are also engineered to express soluble IL-15 to enhance persistence and expansion.
