ILT4.4-1BB.DAP10.TLR2 CIR-NK is an engineered natural killer (NK) cell therapy developed by NKILT Therapeutics. It utilizes a Chimeric ILT Receptor (CIR) platform where the extracellular domain of the Immunoglobulin-like transcript 4 (ILT4) receptor is fused to a novel intracellular signaling architecture comprising 4-1BB, DAP10, and TLR2 costimulatory domains. This design allows the NK cells to specifically target and be activated by tumor cells expressing Human Leukocyte Antigen G (HLA-G), an immune checkpoint often upregulated in cancers to evade immune surveillance. The cells are further modified to express soluble IL-15 for enhanced persistence and a ΔCD19 marker. Preclinical studies have demonstrated potent activity against HLA-G-positive multiple myeloma and acute myeloid leukemia (AML) cells.
