Overview

ILT4.4-1BB.zeta CIR-NK is an experimental chimeric ILT receptor (CIR) engineered natural killer (NK) cell therapy developed by NKILT Therapeutics. It is designed to target HLA-G, a non-classical MHC class I molecule frequently overexpressed in various cancers to evade immune detection. The CIR construct consists of the extracellular domain of the ILT4 (Immunoglobulin-Like Transcript 4) receptor fused to intracellular 4-1BB and CD3-zeta (ζ) signaling domains. This configuration allows the NK cells to recognize HLA-G on tumor cells and trigger cytotoxic activation. In preclinical studies, it has been investigated for the treatment of HLA-G-positive malignancies, including acute myeloid leukemia (AML) and multiple myeloma, though it often serves as a benchmark for newer generations of CIR-NK cells utilizing different costimulatory domains like DAP10.
