IM-1021 is a ROR1-targeted antibody-drug conjugate (ADC) developed by Immunome for the treatment of advanced B-cell lymphomas and solid tumors. The drug consists of a monoclonal antibody directed against receptor tyrosine kinase-like orphan receptor 1 (ROR1), conjugated via a cleavable linker to HC74, Immunome’s proprietary camptothecin-derived DNA topoisomerase I (TOP1) inhibitor payload. This design aims to maximize internalization and the bystander effect in tumor cells with variable ROR1 expression. IM-1021 is currently being evaluated in Phase 1 clinical trials for safety, tolerability, pharmacokinetics, and preliminary anti-tumor activity in patients with high unmet medical need[1][3][5][6]. Preclinical data have shown robust anti-tumor activity across multiple models including mantle cell lymphoma and triple-negative breast cancer[2][4].
