IM-1340

IM-1340 is a preclinical, potentially first-in-class antibody-drug conjugate (ADC) developed by Immunome for the treatment of multiple solid tumors. The drug consists of a proprietary high-affinity antibody linked via a cleavable, undisclosed linker to HC74, a novel topoisomerase I inhibitor (TOP1i) payload. The target of IM-1340 is an undisclosed tumor-associated receptor with unique expression across neuroendocrine tumors (NETs) and various carcinomas—including small cell lung cancer (SCLC), non-small cell lung cancer (NSCLC), pancreatic, and prostate cancers—while showing limited expression in normal tissues. This target promotes tumor growth by accelerating proliferation, cell cycle progression, and migration of cancer cells; it also functions as an endocytic transport receptor for an endolysosomal protease, facilitating efficient ADC internalization. Preclinical studies have demonstrated robust anti-tumor activity with regression observed after single doses in several in vivo models. IND-enabling work began in late 2024[3][5][6].
