Overview

IMA203CD8 is a next-generation, autologous T cell receptor-engineered (TCR-T) cell therapy developed to target solid tumors expressing PRAME (Preferentially Expressed Antigen in Melanoma). The therapy involves the genetic engineering of a patient's own T cells to express both a PRAME-specific TCR and the CD8αβ co-receptor, enabling engagement of both CD4+ and CD8+ T cells. This dual approach is designed to enhance anti-tumor activity by increasing cytotoxicity through CD8+ responses and supporting helper functions via activated CD4+ T cells. IMA203CD8 specifically recognizes HLA-A*02:01-presented PRAME peptides on tumor cells, leading to targeted tumor cell destruction. It is being developed by Immatics for use in patients with recurrent or refractory solid cancers who are HLA-A*02:01 positive and have exhausted standard treatments. The drug is currently in Phase 1 clinical trials for multiple solid tumor indications[1][2][3][5][6][7].
