Overview

IMA203CD8 GEN2 is a second-generation, autologous T-cell receptor (TCR) engineered T-cell therapy developed by Immatics. It is designed to target the PRAME (PReferentially expressed Antigen in MElanoma) antigen, a cancer-testis antigen highly expressed in various solid tumors. Unlike the first-generation IMA203, this GEN2 version involves the co-transduction of T cells with both a PRAME-specific TCR and a CD8αβ co-receptor. This modification is intended to functionalize CD4+ T cells, enabling them to contribute directly to the anti-tumor response alongside CD8+ T cells, thereby enhancing the overall potency and durability of the therapy. It is currently being evaluated in Phase 1 clinical trials for advanced solid tumors, including melanoma, ovarian cancer, and uterine cancer, specifically in patients who are HLA-A*02:01 positive.
