Overview

IMA401 is a bispecific T cell engaging receptor molecule (TCER®) developed by Immatics. It is designed to target an HLA-A*02-presented peptide derived from the cancer-associated proteins melanoma-associated antigen 4 and/or 8 (MAGEA4/8), which are highly prevalent in several solid tumor types including squamous non-small-cell lung carcinoma, head and neck squamous cell carcinoma, bladder, uterine, esophageal and ovarian carcinomas, as well as melanoma and sarcoma subtypes. The TCER® format features two binding regions—one with an affinity-optimized TCR that binds specifically to the tumor target presented by HLA molecules on cancer cells, and another derived from an antibody domain that recruits endogenous T cells for activation against the tumor. This design enables broad activation of T cells regardless of their intrinsic specificity. Preclinical studies have shown anti-tumor activity with complete remissions in various in vivo models. IMA401 is currently being evaluated in Phase 1 clinical trials for patients with recurrent or refractory solid tumors[1][3][4][5].
