Overview

IMA402 is a next-generation, half-life extended bispecific T cell-engaging receptor molecule (TCER) developed to target PRAME (Preferentially Expressed Antigen in Melanoma), an intracellular cancer-testis antigen presented on HLA-A*02:01. It is designed to redirect and activate T cells against PRAME-positive tumor cells by simultaneously binding to the CD3 complex on T lymphocytes and the PRAME peptide-HLA complex on tumor cells. This dual targeting results in selective tumor cell killing with enhanced anti-tumor activity and reduced toxicity compared to earlier T cell engagers. Preclinical studies have shown potent activity against PRAME-positive tumors, favorable pharmacokinetics due to its half-life extension, and a promising safety profile. The drug is currently being evaluated in Phase 1/2 clinical trials for patients with advanced or refractory solid tumors[1][2][3][4][5][7].
