IMA403 is an investigational, preclinical-stage T-cell engaging receptor (TCER®) bispecific molecule developed by Immatics. It is designed to treat solid tumors by targeting a specific peptide derived from the cancer-testis antigens MAGEA3 (Melanoma-associated antigen A3) and MAGEA6 (Melanoma-associated antigen A6), which are presented on the cell surface by the HLA-A*01 major histocompatibility complex. The IMA403 molecule features a soluble, high-affinity T-cell receptor (TCR) domain that recognizes the MAGEA3/A6-HLA complex with high specificity, linked to an anti-CD3 single-chain antibody fragment (scFv). This bispecific architecture allows IMA403 to bridge T cells to tumor cells, triggering T-cell activation and subsequent lysis of the malignant cells. MAGEA3 and MAGEA6 are ideal targets due to their high expression in various cancers—including non-small cell lung cancer (NSCLC), head and neck squamous cell carcinoma (HNSCC), and melanoma—and their restricted expression in normal tissues. IMA403 represents an expansion of the TCER® pipeline to address patient populations with the HLA-A*01 haplotype.
