IMAB027 (also known as ASP1650) is a chimeric monoclonal antibody that targets Claudin 6 (CLDN6), an embryonic tight junction membrane protein aberrantly expressed in several human cancers, particularly ovarian and testicular cancers. CLDN6 is largely absent from normal adult tissues, making it a promising cancer-specific target. IMAB027 exerts its anti-tumor effects primarily through antibody-dependent cellular cytotoxicity and complement-dependent cytotoxicity, leading to apoptosis of CLDN6-positive tumor cells. The drug was developed by Ganymed Pharmaceuticals and later by Astellas Pharma for the treatment of advanced ovarian cancer, testicular cancer, and germ cell tumors. Clinical development included first-in-human Phase I/II trials in patients with recurrent advanced ovarian cancer; however, development has been discontinued for all indications[1][2][3][4][5].
