Drug intelligence / Profile preview
Imanixil (also known as HOE-402 or SAR088) is a small molecule drug originally developed by Sanofi (formerly Sanofi-Aventis / Hoechst). It was initially investigated as a potent cholesterol-lowering and anti-atherosclerotic agent that acts as an inducer of the low-density lipoprotein receptor (LDLR) and inhibits very low-density lipoprotein (VLDL) production. Later, Sanofi characterized it as a selective phosphatidylinositol 5-phosphate 4-kinase beta (PI5P4Kβ, encoded by PIP4K2B) inhibitor with some activity against PI5P4Kα. In preclinical models, it has been shown to lower blood glucose levels in diabetic animal models and enhance autophagosome-lysosome fusion. Additionally, in research settings, it is used as a tool to disrupt lipid trafficking and membrane-dependent endocytosis, which has been shown to impair cell-free hemoglobin clearance in sickle cell disease models. Development of the drug was discontinued in Phase 2.
