Overview

Imatinib + hydroxyurea is a fixed-drug combination of two oral small molecule antineoplastic agents primarily investigated in patients with recurrent or progressive cancers, particularly glioblastoma multiforme and chronic myeloid leukemia. **Imatinib** is a selective tyrosine kinase inhibitor, principally inhibiting BCR-ABL, platelet-derived growth factor receptor (PDGFR), and c-KIT, blocking proliferation of malignant cells dependent on these kinases. **Hydroxyurea** is a ribonucleotide reductase inhibitor that suppresses DNA synthesis, thereby inhibiting the proliferation of rapidly dividing cells. The combination has been studied for potential synergistic antitumor effects, though trials have not demonstrated clear superiority over hydroxyurea monotherapy in recurrent glioblastoma. Both drugs are taken orally.
