Overview

IMB-201 is an investigational antibody-drug conjugate (ADC) developed by IMBiologics that targets human leukocyte antigen-G (HLA-G), a non-classical MHC class I molecule. HLA-G is an immune checkpoint molecule primarily expressed in the placenta to maintain fetal-maternal tolerance, but it is frequently upregulated in various cancers to facilitate immune evasion. By binding to HLA-G, IMB-201 is designed to disrupt the interaction between tumor cells and inhibitory receptors such as ILT2 and ILT4 on T cells, NK cells, and macrophages, thereby reversing tumor-mediated immunosuppression. Furthermore, as an ADC, IMB-201 utilizes a monoclonal antibody to selectively deliver a cytotoxic payload to HLA-G-expressing tumor cells, combining targeted chemotherapy with immunotherapy. Due to the restricted expression of HLA-G in normal adult tissues, IMB-201 is expected to have a favorable safety profile with minimal off-target effects.
