IMB5036 is a **novel pyridazinone compound** with strong antitumor and antiproliferative activity, primarily investigated for its efficacy against pancreatic cancer and hepatocellular carcinoma. Mechanistically, IMB5036 induces **necroptosis** by activating the RIPK1/RIPK3/MLKL pathway, as demonstrated by increased phosphorylation and translocation of MLKL and related proteins in target cells[1][4]. It also partially activates **apoptosis and pyroptosis**, binding directly to KH-type splicing regulatory protein (KSRP), and inducing **gasdermin E (GSDME)-dependent pyroptosis**. IMB5036 overcomes resistance to multiple chemotherapeutic drugs by downregulating P-glycoprotein (P-gp) and is not a substrate of P-gp[7].
