Overview

IMC-002 + rituximab is an investigational combination therapy consisting of **IMC-002**, a next-generation fully human monoclonal antibody targeting **CD47**, and **rituximab**, a chimeric monoclonal antibody targeting **CD20**. IMC-002 is designed to block the CD47 "don't eat me" signal, thereby enhancing the phagocytosis of tumor cells by macrophages and augmenting anti-tumor immunity[2][4]. Rituximab induces immune-mediated killing of B cells, primarily through CDC (complement-dependent cytotoxicity), ADCC (antibody-dependent cellular cytotoxicity), and direct induction of apoptosis[1][5]. The combination is under clinical investigation for the treatment of various hematologic malignancies, especially B-cell lymphomas, leveraging complementary mechanisms: IMC-002 disrupts tumor immune evasion, while rituximab directly depletes CD20+ B cells. IMC-002 is being developed by ImmuneOncia Therapeutics, with clinical manufacturing supported by Samsung Biologics[2]. Rituximab is marketed by Roche/Genentech and Biogen.
