Overview

IMC-EB10 is a fully human IgG1 monoclonal antibody that targets the Fms-like tyrosine kinase 3 receptor (FLT3, also known as CD135), a class III receptor tyrosine kinase highly expressed in acute myeloid leukemia (AML) and some acute lymphoblastic leukemia (ALL) cells. By binding with high affinity to FLT3, IMC-EB10 blocks the interaction between FLT3 and its ligand, inhibiting downstream signaling pathways such as MAPK and AKT that are critical for leukemic cell proliferation and survival. The antibody demonstrates both antiproliferative effects in vitro and efficacy in mouse models of AML and ALL by prolonging survival and reducing leukemic engraftment. Its mechanism includes direct inhibition of FLT3 phosphorylation as well as potential antibody-dependent cell-mediated cytotoxicity (ADCC). Developed originally by ImClone Systems (a subsidiary of Eli Lilly), clinical development reached phase 1 trials for relapsed/refractory AML but was discontinued[1][2][4][5][6][7][8].
