IMC-KRASG12D is a soluble bispecific ImmTAC (Immune mobilizing monoclonal TCR Against Cancer) molecule designed to target the KRAS G12D neoantigen. Developed by Immunocore, it consists of an affinity-enhanced T cell receptor (TCR) specific for the KRAS G12D decamer peptide (VVVGADGVGK) presented by HLA-A*11:01, fused to a humanized anti-CD3 single-chain variable fragment (scFv). The molecule works by redirecting and activating polyclonal T cells to selectively kill cancer cells harboring the KRAS G12D mutation, a common driver in pancreatic and colorectal cancers, while sparing cells expressing wild-type KRAS. Preclinical studies have demonstrated its high specificity and potency in mediating T-cell activation and redirected killing of KRAS G12D-positive tumor cell lines.
