IMC-R117C is a bispecific T cell receptor (TCR) immunotherapy developed using Immunocore’s ImmTAC (Immune Mobilizing Monoclonal TCRs Against Cancer) platform. It targets PIWIL1, a cancer-testis antigen overexpressed in multiple malignancies including colorectal and other gastrointestinal cancers. The molecule consists of a high-affinity monoclonal TCR specific for PIWIL1 presented by HLA-A*02:01 on tumor cells, fused to an anti-CD3 effector domain that recruits and activates T cells to kill the targeted cancer cells. This design enables the immune system to recognize and destroy tumor cells expressing intracellular antigens not accessible by conventional antibody therapies. IMC-R117C is currently being evaluated in Phase 1/2 clinical trials as both monotherapy and in combination with standard-of-care treatments for advanced solid tumors expressing PIWIL1[2][3][5][7].
