IMC-U120AI is a bispecific, recombinant fusion protein designed as a CD1a-tethered PD1 agonist, developed by Immunocore. It is the first non-HLA-restricted (universal) candidate from Immunocore’s ImmTAAI platform, intended for tissue-specific down-modulation of the immune system in autoimmune diseases. The drug targets CD1a, an HLA-like protein expressed on skin and mucosal antigen-presenting cells such as Langerhans cells, which play a key role in allergic inflammation seen in atopic dermatitis and potentially other immune-mediated diseases. Its dual mechanism of action involves blocking CD1a to suppress activation by lipid antigens and activating PD1 on T cells to suppress activation by peptide antigens. The initial indication under development is atopic dermatitis[1][3][5][6].
