Overview

NY-ESO-ImmTAC (IMCny1) is a bispecific soluble T-cell receptor (TCR) fusion protein developed by Immunocore and formerly partnered with GSK (as GSK3562490). It belongs to the ImmTAC (Immune-mobilizing monoclonal TCRs Against Cancer) class of biologics, designed to overcome the limitations of monoclonal antibodies by targeting intracellular antigens. The molecule consists of a high-affinity TCR engineered to recognize the NY-ESO-1 (New York esophageal squamous cell carcinoma 1) peptide SLLMWITQC presented by HLA-A*02:01, fused to an anti-CD3 single-chain variable fragment (scFv). By binding both the tumor-specific peptide-MHC complex and the CD3 receptor on T cells, IMCny1 redirects non-specific T cells to kill NY-ESO-1-positive tumor cells. Clinical development reached Phase 1 for indications such as synovial sarcoma and non-small cell lung cancer before being discontinued by GSK in 2021.
