Overview

IMCnyeso is a bispecific immune mobilizing monoclonal T cell receptor against cancer (ImmTAC) molecule developed to treat advanced cancers by redirecting the body's own immune system. It consists of a soluble affinity-enhanced T-cell receptor (TCR) specific for human leukocyte antigen A*02:01 (HLA-A*02:01)-restricted cancer-testis antigens NY‑ESO‑1 and LAGE‑1A fused to an anti-CD3 single-chain variable fragment. This design enables the drug to target tumor cells expressing these antigens and recruit cytotoxic T cells for targeted killing. IMCnyeso was primarily investigated in patients with advanced solid tumors such as non-small cell lung cancer (NSCLC), melanoma, urothelial carcinoma (bladder cancer), and synovial sarcoma who are HLA-A*02:01 positive with tumors expressing NY‑ESO‑1 or LAGE‑1A[2][5][6]. The drug was administered as weekly intravenous infusions in clinical trials but development has been discontinued[2][6].
