IMD-2358 is a tumor microenvironment-activated antibody-drug conjugate (TMEA-ADC) targeting receptor tyrosine kinase-like orphan receptor 1 (ROR1). Developed by Shanghai Affinity Biopharmaceutical, the drug utilizes the company's proprietary TMEA platform, which is designed to improve the therapeutic index by ensuring the payload is released specifically within the tumor microenvironment. IMD-2358 incorporates a DXd (deruxtecan) payload, a potent topoisomerase I inhibitor. ROR1 is an onco-embryonic antigen highly expressed in various solid tumors but with limited expression in normal adult tissues, making it an attractive target for ADC therapy. The drug is currently being investigated for the treatment of advanced solid tumors.
