IMD-2816 is a bispecific T-cell engager (TCE) developed by Shanghai Affinity Biopharmaceutical for the treatment of solid tumors, particularly those of gastrointestinal origin. The drug is engineered using the company's proprietary Tumor Microenvironment Activated (TMEA) platform, which is designed to improve the safety and therapeutic window of T-cell engagers. IMD-2816 simultaneously targets Cadherin-17 (CDH17), a cell adhesion protein frequently overexpressed in colorectal, gastric, and pancreatic cancers, and the CD3 receptor on T cells. By cross-linking these targets, the drug facilitates the recruitment and activation of cytotoxic T cells directly to the tumor site, leading to the selective destruction of CDH17-positive cancer cells. The TMEA technology incorporates a masking or conditional activation mechanism intended to keep the drug inactive in systemic circulation, thereby minimizing off-tumor toxicities and cytokine release syndrome (CRS) while allowing for potent activation within the protease-rich tumor microenvironment.
