Overview

IMD-4482 is a novel, orally administered small molecule inhibitor targeting plasminogen activator inhibitor-1 (PAI-1). It was developed to inhibit the function of PAI-1, which is implicated in cancer progression and poor prognosis, particularly in ovarian cancer. Preclinical studies have shown that IMD-4482 induces G0/G1 cell cycle arrest and inhibits proliferation of PAI-1-positive ovarian cancer cells. Mechanistically, it disrupts cell adhesion to vitronectin by interfering with the PAI–urokinase receptor complex and integrin αVβ3 interaction, leading to reduced phosphorylation of extracellular signal-regulated kinase (ERK) and focal adhesion kinase (FAK). In xenograft mouse models, IMD-4482 significantly inhibited peritoneal dissemination of ovarian cancer cells[1][9][10]. The drug is being explored for its potential in treating ovarian cancer as well as neurodegenerative diseases such as Alzheimer's disease and cerebrovascular dementia[7].
