Overview

imExo (immunogenic exosomes) are engineered extracellular vesicles developed for targeted cancer immunotherapy, specifically for the treatment of glioblastoma. Developed by researchers at MD Anderson Cancer Center, these exosomes are loaded with interferon-gamma (IFN-γ) mRNA using a microfluidic electroporation approach. The surface of the exosomes is engineered to overexpress CD64 (FcγRI), which serves as a modular adaptor to dock various targeting ligands, such as anti-CD71 or anti-PD-L1 antibodies. This design allows the exosomes to selectively target tumor cells and cross biological barriers like the blood-brain barrier. Once delivered, the IFN-γ mRNA cargo induces potent antitumor activity and can overcome intrinsic resistance to standard immunotherapies in preclinical models.
