Overview

IMG-004 is a potent, highly selective, noncovalent, reversible and brain-permeable small molecule inhibitor of Bruton's tyrosine kinase (BTK). It is designed specifically for inflammatory and autoimmune diseases that require long-term treatment. The drug has demonstrated a favorable safety profile in phase 1 studies with healthy adults, showing an extended half-life (26–37 hours), robust target engagement (up to 95% inhibition of basophil activation at certain doses), and durable pharmacodynamic effects that may allow for once-daily oral dosing. Originally discovered by HUTCHMED and now developed by Inmagene Biopharmaceuticals, IMG-004 is being evaluated for chronic spontaneous urticaria (CSU), rheumatoid arthritis, hidradenitis suppurativa, and multiple sclerosis
