IMG-007 is a humanized, non-depleting anti-OX40 IgG1 monoclonal antibody engineered with a silenced antibody-dependent cellular cytotoxicity (ADCC) function and an extended half-life. It specifically blocks the OX40/OX40L signaling pathway in both blood and tissues without depleting T cells, aiming to minimize safety risks while maintaining efficacy. The drug is being developed for immune-mediated inflammatory diseases, particularly moderate-to-severe atopic dermatitis (AD) and severe alopecia areata (AA). In clinical trials, IMG-007 has demonstrated durable inhibition of serum inflammatory markers associated with Th1, Th2, Th17, and CD8+ T cells. It is administered intravenously or subcutaneously; the subcutaneous formulation has shown a mean terminal half-life of approximately 31–35 days. Clinical studies have reported robust efficacy signals—such as significant reductions in EASI scores for AD and SALT scores for AA—with a favorable safety profile characterized by no serious adverse events or treatment discontinuations[2][3][4][5][6][7][8].
