Overview

IMGN2293 is an orally available, highly selective small molecule inhibitor of Tyrosine Kinase 2 (TYK2), a member of the Janus kinase (JAK) family. By selectively targeting the pseudokinase (JH2) domain of TYK2, the drug modulates signaling pathways for key pro-inflammatory cytokines, including interleukin-23 (IL-23), interleukin-12 (IL-12), and Type I interferons, which are central to the pathogenesis of various autoimmune and inflammatory disorders. Developed by Inmagene Biopharmaceutics (which entered a merger agreement with Ikena Oncology in late 2024), IMGN2293 is being advanced into Phase 3 clinical trials for the treatment of hidradenitis suppurativa (HS) and psoriatic arthritis (PsA). Its high selectivity for TYK2 over other JAK family members (JAK1, JAK2, and JAK3) is intended to provide a superior safety profile by avoiding off-target effects such as hematological abnormalities and lipid elevations commonly associated with pan-JAK inhibition.
