IMGN388

IMGN388 is an investigational antibody-drug conjugate (ADC) composed of a fully human monoclonal antibody targeting alpha-v integrins conjugated to the cytotoxic maytansinoid agent DM4. The drug binds to integrins expressed on the surface of tumor cells and endothelial cells involved in angiogenesis. Upon binding and internalization by target cells, the DM4 payload is released intracellularly where it binds tubulin and disrupts microtubule assembly/disassembly dynamics, leading to inhibition of cell division and tumor growth. IMGN388 was developed for the treatment of various solid tumors including lung, colon, pancreatic, ovarian, breast, prostate, neuroendocrine and uterine cancers. It demonstrated preclinical efficacy in xenograft models but development was discontinued after phase I clinical trials[1][2][3][4][6].
