Imifoplatin (PT-112) is a novel platinum-pyrophosphate chemotherapy agent under investigation primarily for the treatment of advanced solid tumors, including metastatic castration-resistant prostate cancer (mCRPC) and non-small cell lung cancer (NSCLC). Its mechanism of action involves inducing immunogenic cell death, which not only directly kills tumor cells but also stimulates an immune response against the cancer. The presence of pyrophosphate in its structure facilitates high localization to bone tissue, making it particularly active against bone metastases. Clinical trials have shown that imifoplatin monotherapy and combination therapy with immune checkpoint inhibitors like avelumab are well tolerated and demonstrate therapeutic activity, including reduction in serum alkaline phosphatase (ALP), improvement in bone pain, and disease control in heavily pretreated patients.
