Overview

IMM-1-104 + gemcitabine + nab-paclitaxel is a combination therapy under investigation for the treatment of advanced solid tumors, particularly pancreatic cancer. IMM-1-104 is an oral small molecule MEK inhibitor that selectively binds to and inhibits MEK1/2 activity. This inhibition blocks downstream signaling in the MAPK pathway, which can suppress tumor cell proliferation and growth factor-mediated signaling[1][5]. The drug has shown promising results in early clinical trials for pancreatic cancer when combined with modified regimens of gemcitabine (a nucleoside analog chemotherapy) and nab-paclitaxel (an albumin-bound formulation of paclitaxel), both standard chemotherapies that disrupt cell division by targeting rapidly dividing cells[6][7]. The combination aims to enhance antitumor efficacy by targeting both the MAPK pathway (with IMM-1-104) and cellular replication mechanisms (with chemotherapy). Immuneering is developing IMM-1-104; Gemzar (gemcitabine) is developed by Eli Lilly; Abraxane (nab-paclitaxel) was developed by Abraxis BioScience/Celite but is now marketed by Bristol Myers Squibb.
