IMM-4701 (also referred to as CD24 mAb-CD47 trap) is a preclinical-stage bispecific antibody-trap fusion protein developed by ImmuneOnco. It is constructed using a humanized anti-CD24 monoclonal antibody (IgG1) backbone conjugated to a SIRPα decoy receptor at the N-terminal of the antibody heavy chain. By simultaneously targeting and neutralizing both CD24 (blocking the CD24/Siglec-10 pathway) and CD47 (blocking the CD47/SIRPα pathway), IMM-4701 acts as a dual 'don't eat me' checkpoint inhibitor. This dual blockade enhances macrophage-mediated phagocytosis and antitumor immune responses. Preclinical studies have demonstrated significant tumor growth inhibition in models of triple-negative breast cancer and Herceptin-resistant breast cancer.
