IMM20059 is a preclinical-stage monoclonal antibody developed by Immunome that targets the N-terminal domain of epsin 1 (EPN1). EPN1 is an adapter protein typically involved in clathrin-mediated endocytosis and is usually located intracellularly; however, it becomes ectopically expressed on the surface of various tumor cells, including melanoma, lung, breast, and prostate cancers. By targeting this tumor-specific surface expression, IMM20059 enables selective tumor targeting. Preclinical data suggests that IMM20059 can enhance the efficacy of immune checkpoint inhibitors like atezolizumab by increasing the production of intratumoral chemokines such as MIP-1α, MIP-1β, and RANTES, potentially converting "cold" tumors into "hot" tumors more susceptible to immunotherapy.
