IMM20253 is a patient-derived human monoclonal antibody developed by Immunome for the treatment of COVID-19. It is a component of the IMM-BCP-01 antibody cocktail, which also includes IMM20184 and IMM20190. IMM20253 targets a cryptic epitope on the outer surface of the receptor-binding domain (RBD) of the SARS-CoV-2 spike protein. Unlike many other COVID-19 antibodies, it operates via an ACE2-independent mechanism; binding to its epitope alters the conformation of the spike trimer and promotes the release of spike monomers, thereby neutralizing the virus. The development of this antibody was supported by the U.S. Department of Defense to provide a broad-spectrum therapeutic effective against various SARS-CoV-2 variants of concern.
