IMMU-114 is a humanized IgG4 monoclonal antibody that targets the human leukocyte antigen HLA-DR, a member of the MHC class II antigen family expressed on various hematologic and some solid tumors. Unlike earlier anti-HLA-DR antibodies, IMMU-114 is engineered to lack effector-cell functions such as complement-dependent cytotoxicity (CDC) and antibody-dependent cellular cytotoxicity (ADCC), reducing toxicity and infusion reactions. Its antitumor activity is mediated through direct binding to HLA-DR, leading to hyperactivation of ERK and JNK mitogen activated protein kinase signaling pathways in malignant cells, which induces apoptosis independently of immune effector mechanisms. IMMU-114 has shown preclinical efficacy against mantle cell lymphoma, acute lymphoblastic leukemia (ALL), hairy cell leukemia (HCL), chronic lymphocytic leukemia (CLL), multiple myeloma (MM), non-Hodgkin lymphoma (NHL) including rituximab-resistant cases, with limited effect in acute myeloid leukemia despite high HLA-DR expression. It has been evaluated in phase I clinical trials for relapsed or refractory NHL and CLL[1][3][5][6][7].
