Drug intelligence / Profile preview
ImmuneNanoVaccine Hydrogel is an injectable, thermo-responsive hydrogel-based cancer vaccine designed for the treatment of BCG-resistant bladder cancer. It is co-encapsulated with programmed death-ligand 1 (PD-L1) blockades (anti-PD-L1 antibodies) and polymeric nanoparticles loaded with three immunostimulatory agonists targeting Toll-like receptor 8 (TLR8) and the stimulator of interferon genes (STING) pathway. Upon intravesical administration, the hydrogel undergoes a rapid sol-to-gel transformation at body temperature, which prolongs tumor retention, sustains the release of the immunotherapeutic agents, and reduces acute systemic inflammation. In preclinical orthotopic bladder mouse models, the hydrogel has demonstrated significant antitumor immunity by increasing CD8+ T cells and memory T cells while reducing myeloid-derived suppressor cells and M2-like macrophages in the tumor microenvironment.
