Immuno-STAT (IST) is a dimeric protein scaffold designed to selectively activate and expand antigen-specific CD8+ T cells. It functions by delivering peptide-specific T-cell receptor (TCR) activation, either alone or in combination with CD28 costimulatory signals. This platform aims to overcome logistical challenges associated with conventional adoptive cell transfer methods by providing a scalable modality for generating polyfunctional cytotoxic CD8+ T cells. Immuno-STAT has demonstrated the ability to expand naive T cells targeting HIV-associated SL9 or melanoma-associated MART-1 epitopes, showing potential for treating HIV, other persistent viral infections, and cancer.
