Overview

Immunoglobulin G (IgG) ATAC degraders are experimental small molecule extracellular protein degraders developed by Avilar Therapeutics. Utilizing the ASGPR Targeting Chimera (ATAC) platform, these bifunctional molecules consist of a high-affinity ligand for the asialoglycoprotein receptor (ASGPR) linked to a binder for the target protein, in this case, immunoglobulin G. By binding both ASGPR on hepatocytes and circulating IgG, the ATAC shuttles the antibody to the endolysosome for degradation. Preclinical data in non-human primates demonstrated significant reduction (up to 85%) of circulating IgG following intravenous or subcutaneous administration. This approach is intended to treat autoimmune and other diseases driven by pathogenic IgG antibodies.
