MSC-IPr is a preclinical cellular vaccine platform developed by CELL Technologies. It consists of mesenchymal stromal cells (MSCs) genetically engineered to express the three inducible subunits of the murine immunoproteasome (β1i, β2i, and β5i) separated by viral T2A sequences. This modification transforms the MSCs into efficient antigen-presenting cells capable of cross-presenting a diverse repertoire of epitopes. The therapy enhances major histocompatibility complex (MHC) class I and CD80 expression, induces de novo production of interleukin-12 (IL-12), and increases chemokine secretion. In preclinical mouse models of EL4 and A20 lymphoma and B16 melanoma, MSC-IPr has demonstrated the ability to reactivate T cell immunity and control tumor growth, particularly when combined with immune checkpoint inhibitors like anti-PD-1 or anti-CTLA4.
