Drug intelligence / Profile preview
Imneskibart + aldesleukin + avelumab is an investigational triplet immunotherapy regimen being evaluated for the treatment of advanced solid tumors, particularly checkpoint inhibitor-refractory adenocarcinoma non-small cell lung cancer (NSCLC). The regimen combines imneskibart (AU-007), a human IgG1 monoclonal antibody designed using artificial intelligence to selectively bind interleukin-2 (IL-2) and prevent its interaction with the CD25 (IL-2Rα) subunit; aldesleukin (Proleukin), a recombinant IL-2 administered at a low subcutaneous loading dose; and avelumab (Bavencio), an anti-PD-L1 monoclonal antibody with active Fc-mediated antibody-dependent cellular cytotoxicity (ADCC) effector function. By blocking IL-2 binding to CD25, imneskibart prevents the expansion of immunosuppressive regulatory T cells (Tregs) and avoids high-dose IL-2 toxicities like vascular leak syndrome, while redirecting both endogenous and exogenous IL-2 to expand and activate CD8+ effector T cells and natural killer (NK) cells. The increased NK cell population in turn enhances avelumab-mediated ADCC against tumor cells, creating a synergistic anti-tumor immune response.
