Overview

IMO-2055 is a synthetic immunomodulatory oligonucleotide and a Toll-like receptor 9 (TLR9) agonist. It mimics unmethylated CpG sequences found in bacterial DNA to stimulate the innate immune system via TLR9 activation. This immune stimulation is intended to enhance antitumor responses and potentiate the efficacy of other anticancer agents. Developed primarily as an anticancer agent for solid tumors such as colorectal cancer, non-small cell lung cancer (NSCLC), head and neck cancer, and renal cell carcinoma, it was investigated in combination with various chemotherapeutic and targeted therapies. Despite promising early-phase results showing immune activation and some disease control in NSCLC trials when combined with agents like Tarceva (erlotinib) and Avastin (bevacizumab), later-stage studies failed to meet primary endpoints for progression-free survival in several cancers. Development has been discontinued or suspended for all major indications[2][4][6][7][8].
