IMP-288 is a synthetic hapten-peptide designed for use in pretargeted radioimmunotherapy and molecular imaging of cancer. It contains a di-HSG (histamine-succinyl-glycine) motif and can be radiolabeled with various isotopes, including ^111In, ^90Y, ^177Lu, ^68Ga, and ^124I. The drug is administered after a bispecific monoclonal antibody (bsMAb) that targets both tumor antigens (such as CEA or Trop-2) and the HSG motif; this allows for highly specific delivery of the radiolabeled peptide to tumor cells while minimizing exposure to normal tissues. IMP-288 has been investigated primarily in solid tumors such as metastatic colorectal carcinoma, non-small cell lung cancer, small cell lung cancer, and advanced colorectal adenocarcinoma. Its mechanism involves acting as a hapten that binds to pre-localized bsMAbs on tumor cells for targeted radionuclide therapy or imaging[1][2][5][6][7].
