IMP13 is a small molecule inhibitor of ubiquitin-specific-processing protease 1 (USP1) developed by Impakt Therapeutics. USP1 is a deubiquitinase that plays a critical role in the DNA damage response (DDR) pathway by regulating the monoubiquitination of proteins such as PCNA and FANCD2, which are essential for translesion synthesis and the Fanconi anemia pathway. By inhibiting USP1, IMP13 exploits synthetic lethality in tumors with existing DNA repair deficiencies, particularly those with BRCA1/2 mutations. It is being investigated for the treatment of various cancers, including those that are resistant to platinum-based chemotherapy or PARP inhibitors. The drug is currently in early-stage clinical development.
