Drug intelligence / Profile preview
IMP1734 + abiraterone is an investigational combination regimen consisting of IMP1734 (also known as EIK1003), an oral, highly selective small-molecule inhibitor of poly(ADP-ribose) polymerase 1 (PARP1), and abiraterone (often administered as abiraterone acetate), an approved small-molecule inhibitor of cytochrome P450 17A1 (CYP17A1) that blocks androgen biosynthesis. Developed through a collaboration between Impact Therapeutics and Eikon Therapeutics, the combination is designed to exploit synthetic lethality in tumors with homologous recombination repair (HRR) gene alterations while minimizing the hematological toxicities typically associated with non-selective PARP1/2 inhibitors. The regimen is being evaluated in a Phase 1/2 clinical trial (NCT06253130) for the treatment of patients with advanced or metastatic prostate cancer, including metastatic castration-resistant prostate cancer (mCRPC) and metastatic castration-sensitive prostate cancer (mCSPC).
