Overview

IMP22 is a preclinical-stage small molecule dual inhibitor of PKMYT1 (Protein Kinase, Membrane Associated Tyrosine/Threonine 1) and WEE1 kinase, developed by Impact Therapeutics. Both PKMYT1 and WEE1 are critical regulators of the G2/M cell cycle checkpoint, acting by phosphorylating and inactivating CDK1 to prevent premature entry into mitosis. By simultaneously inhibiting these two kinases, IMP22 is designed to drive cancer cells into premature mitosis, leading to mitotic catastrophe, DNA damage, and cell death. This mechanism is particularly effective in tumors with specific genetic vulnerabilities, such as CCNE1 amplification or other DNA damage response (DDR) deficiencies. The compound is currently being investigated for the treatment of advanced solid tumors.
