Overview

IMP305 is an ultrasound-sensitive liposomal formulation of exatecan, a potent topoisomerase I inhibitor, specifically engineered for the treatment of pancreatic ductal adenocarcinoma (PDAC). The formulation is designed to address the therapeutic challenges posed by the dense stromal architecture of PDAC, which typically hinders drug penetration. IMP305 utilizes ultrasound-mediated cavitation to trigger localized drug release and physically disrupt the tumor stroma, thereby enhancing intratumoral accumulation. Preclinical studies have shown that IMP305 significantly increases the maximum tolerated dose of exatecan and, when combined with focused ultrasound, demonstrates superior antitumor efficacy and survival rates in pancreatic cancer models compared to free exatecan.
