Drug intelligence / Profile preview
IMP3201 is a preclinical-stage, first-in-class antibody-drug conjugate (ADC) developed by IMPACT Therapeutics. It is designed to target Carcinoembryonic Antigen-Related Cell Adhesion Molecule 5 (CEACAM5), a cell-surface glycoprotein highly overexpressed in various epithelial-derived malignant tumors. IMP3201 is uniquely engineered using IMPACT's proprietary branched linker dual-payload ADC technology platform, delivering two distinct payloads: exatecan, a topoisomerase I (TOPOi) inhibitor, and a proprietary ATR inhibitor (ATRi) at an optimized molar ratio. By concurrently inducing DNA damage via exatecan and blocking DNA damage repair pathways via the ATR inhibitor, IMP3201 generates strong synergistic anti-tumor effects. This dual-payload mechanism is designed to overcome tumor insensitivity and acquired resistance associated with conventional single-payload TOPOi-based ADCs. The candidate is being developed for the treatment of gastrointestinal malignancies, including colorectal cancer, pancreatic cancer, gastric cancer, and esophageal squamous cell carcinoma.
